A section of the octopine Ti plasmid pTiACH5 including the region always integrated in crown gall tumour cells (the TL-DNA) was cloned into the yeast — E. coli shuttle vector pJDB207. This plasmid, pOTY8, was transformed into S. cerevisiae where transcription of the T-DNA was demonstrated, but there was no evidence for the production of octopine synthetase.
The TL-DNA was cloned into a derivative of pJDB207 unable to replicate in yeast, but the TL-DNA did not appear to contain any autonomously replicating sequences (ars) functional in yeast.
