Abstract

The development of a molecular classification of seed storage prolamins in the post-genomic era provides insights into the relationships between these proteins from different cereal species that were not possible when relying on the traditional classification based on sequential extraction. This article summarizes the sequences, structures and relationships between seed storage prolamins from different cereal species and their potential for driving cross-reactive allergies. The applicability of individual wheat seed storage proteins to improve diagnosis of wheat allergy is discussed, highlighting the importance and limitations of the u5-gliadin allergen Tri a 19 in component resolved diagnosis.
